A production-driven component swap inside a validated autoclave loading pattern would normally have triggered three full penetration studies. A targeted scientific assessment removed the need entirely — saving a full production day and preserving compliance.

The Challenge
A production requirement forced one of the items in a validated autoclave loading pattern to be replaced with a similar component. Under conventional practice, even a like-for-like swap of this type would have triggered three full heat-penetration runs to re-qualify the pattern — consuming a production day, demanding significant Production, Validation, and QA effort, and delaying downstream manufacturing.
The Approach
Arnan performed a documented scientific comparison between the existing and replacement items, evaluated against the parameters that actually govern sterilization performance: flow dynamics, heat-transfer characteristics, and steam behavior. The assessment focused specifically on each item’s impact on air removal from the chamber and on steam penetration into the most difficult-to-access internal locations.
The analysis demonstrated that the replacement item was less restrictive to both air removal and steam flow than the original — making it a less challenging case for the already-validated sterilization cycle. This formed a defensible, scientifically grounded rationale to omit the repeat penetration studies altogether.
The Outcome
- Three full heat-penetration runs eliminated through documented rationale
- A complete production day saved, with manufacturing continuing without interruption
- Significant workload avoided for Production, Validation, and QA
- Full regulatory compliance maintained with scientific justification on record
- Demonstrated how a small, well-reasoned assessment can replace weeks of revalidation
